چکیده

T he evolving concept that coronary artery patency after myocardial infarction is associated with improved shortand long-term survival1'2 and left ventricular function34 has intensified efforts to open thrombosed vessels. The importance of maintaining vessel patency has also become clear. Ohman et a15 found that patients with documented reocclusion had more complicated hospital courses, higher in-hospital mortality rates, and worse infarct zone function. Efforts to maintain patency have been hampered by a significant rate of reocclusion, approximately 12-18% after successful thrombolytic therapy for acute myocardial infarction.6,7 Because this rethrombosis occurs in the setting of a systemic fibrinolytic state, therapeutic efforts have been directed toward inhibition of platelet activation and aggregation. This has focused attention on anticoagulant mechanisms native to the vessel itself.

Cheminformatics-Based Anticoagulant Study of Traditionally Used Medicinal Plants

Background: Medicinal plants, as a complementary medicine, have been used to treat various diseases since ancient times. These plants have numerous beneficial applications and are the source of certain conventional drugs. In diseases such as stroke and ischemia, which are caused by several factors, abnormal coagulation is an important causative factor. Accordingly, novel and effective therapies...

Characterization of the conformational alterations, reduced anticoagulant activity, and enhanced antiangiogenic activity of prelatent antithrombin.

A conformationally altered prelatent form of antithrombin that possesses both anticoagulant and antiangiogenic activities is produced during the conversion of native to latent antithrombin (Larsson, H., Akerud, P., Nordling, K., Raub-Segall, E., Claesson-Welsh, L., and Björk, I. (2001) J. Biol. Chem. 276, 11996-12002). Here, we show that the previously characterized prelatent antithrombin is a ...

Preparation of aminated fucoidan and its evaluation as an antithrombotic and antilipemic agent.

Fucoidan, a sulfated poly(L-fucopyranose), is an effective anticoagulant in vitro and in vivo. In the present study, an aminated derivative of fucoidan was prepared and examined for its fibrinolytic and anticoagulant activities. The aminated derivative was more potent than native fucoidan as a stimulator of tissue plasminogen activator-induced plasma clot lysis, and its effectiveness was compar...

Chemical synthesis and spontaneous folding of a multidomain protein: anticoagulant microprotein S.

Because of recent high-yield native ligation techniques, chemical synthesis of larger multidomain bioactive proteins is rapidly coming within reach. Here we describe the total chemical synthesis of a designed "microprotein S," comprising the gamma-carboxyglutamic acid-rich module, the thrombin-sensitive module, and the first epidermal growth factor-like module of human plasma protein S (residue...
